Peptide CJC-1295 DAC

CJC-1295 is a tetrasubstituted peptide hormone that consists of 30 amino acids. In the human body, it functions as an analogue of somatoliberin (a natural growth hormone secretion stimulator).

One of the main advantages of CJC-1295 DAC compared to the widely used GRF(1-29) analogue (synonyms: CJC-1295 w/o, without or without DAC) is its long half-life (up to two weeks). CJC-1295 DAC binds to plasma proteins (albumin), which leads to a significantly longer duration of action, due to the addition of lysine bound to the non-peptide molecule DAC (Drug Affinity Complex).

Be sure to find out when purchasing if CJC-1295 contains the DAC (Drug Affinity Complex) molecule, this will determine the efficacy and regimen of the course. History

CJC-1295 DAC was invented at the Canadian biotechnology company ConjuChem in 2005, but did not gain popularity in bodybuilding until 2010-2011, when third-party companies, including Chinese manufacturers, took over the production technology. The time of action (half-life) is 6-8 days.

ConjuChem initiated research on CJC-1295 DAC in 2005, the purpose of which was to study the effect of the substance on visceral fat in AIDS and obese patients. It is known that growth hormone had previously been used in similar situations to burn fat. The results were highly successful for most patients, but the research was halted after lawsuits were filed by three patients who suffered a myocardial infarction after a course of CJC-1295 DAC.

Mechanism of Action

CJC-1295 acts on the nuclei of the anterior pituitary lobe and stimulates growth hormone secretion, mainly through the cAMP-dependent pathway. The substance binds to GHRH receptors, which belong to the G-protein coupled receptor group.

Studies have shown that CJC-1295 DAC has similar effects to growth hormone:

A brief overview of the physiology of growth hormone

The triggering of growth hormone (GH) release by the pituitary gland depends on three main types of regulatory hormones:

  • Somatostatin, which suppresses GH secretion, is what provides the pulse-like curve of secretion (creates peaks in concentration at certain hours, particularly at night).
  • Growth hormone releasing hormone (GHRH), is a stimulating release peptide. GHRH administration causes a wave-like increase in concentrations, which will be weak during hours when natural GH secretion is decreased by somatostatin, and high during natural GH elevation (for example, at night). In other words, GHRH increases GH secretion without affecting the natural pulse response. Artificial analogues: GRF(1-29), aka Sermorelin and CJC-1295.
  • Ghrelin (GHRP) is a secretion modulator, i.e. it regulates the balance between the peaks and drops of GH concentration. Artificial analogues are GHRP-6, GHRP-2, Hexarelin, and Ipamorelin. They create a peak in GH concentration immediately after administration, regardless of the time of day or the presence of somatostatin in the blood. As we age, ghrelin helps to maintain growth hormone secretion despite the increasing influence of somatostatin.

CJC-1295 DAC in Bodybuilding

After a single injection of CJC-1295 DAC, the hormone concentration in the blood increases 2-10 times by day 6, the concentration of IGF-I increases 1,5-3 times by day 10. The elimination half-life is 6-8 days. After several injections, the level of insulin-like growth factor-1 (IGF-1) remains elevated for a month. The average dose is 30-60 micrograms per kg body weight, once a week.

There is very frequent disagreement about the frequency of administration of the drug. Thus, the following concentration curves at different dosages have been obtained in studies:

The calculations show that in order to maintain the optimal concentration, it is reasonable to administer the peptide twice a week, because by the fifth day it drops to ineffective. We will also give graphs of the use of different doses of the drug, which show that the optimal concentration of IGF-1 is observed at a dose of 60 mcg/kg (approximately 2,000 mcg, twice a week), while increasing the dose to 125 mcg/kg practically does not increase the concentration of IGF-1.

Thus, the following protocol can be considered optimal: 1000-2000 mcg (1 or 1/2 vial) twice a week. In addition to training, don’t forget to eat a proper diet and take sports nutrition to achieve your goal.

Combined Course

It has been well proven and accurately established that the simultaneous administration of GHRH (e.g., CJC-1295) and GHRP (e.g., GHRP-6 or GHRP-2) has a synergistic effect. These peptides have a potentiating effect on each other. In other words, when CJC-1295 alone is administered, we will get a stimulating effect of 2 points. When administering GHRP-6 alone, we will get a 4-point effect. When administering both substances at the same time we will get an effect not of 6 points (additive effect 2+4=6), as you might expect, but of 10 points – more than the sum of each separately (potentiating effect 2+4=10). In this way we get not only a greater anabolic response, but also can save money.

Although the natural pulsation of GH is preserved when CJC-1295 is administered, it does not cause an increase in these peaks, increasing only the basal GH level, thus creating a female-type concentration pattern with a smaller anabolic response. Moreover, GHRH is only effective when somatostatin levels are low. Therefore, combining CJC-1295 and GHRP-6 or GHRP-2 allows you to maintain a natural curve, and adjust the timing and height of concentration peaks.

Optimal Combination Course:

  • GHRP-6 or GHRP-2 100 mcg three times a day.
  • CJC-1295 DAC at 1000 mcg twice a week.
  • The course duration is 8-12 weeks.

In terms of effectiveness, it is equal to a cycle of growth hormone at 15 IU/day, while the cost is several times lower.

Storage and preparation

According to J. Bombela recommendations, the vial should be cooled to room temperature, then the required volume of water for injection should be introduced (which should flow down the wall of the vial) and stirred by rotating movements (do not shake). The solution obtained with the water for injection remains stable for only 8-10 days in the refrigerator. If bacteriostatic water is used, the storage time is increased to 30 days in the refrigerator. CJC-1295 DAC is best dissolved with bacteriostatic water only. The resulting solution should not be frozen.

CJC-1295 without DAC

The optimal dose of CJC-1295 without DAC: 1 mcg/kg up to three times a day (when injected subcutaneously) (in a vial 2mg = 2000mcg). Thus, at a weight of 90 kg, the relevant dosage would be 90 mcg, and one vial would be sufficient for 8-22 days (depending on the frequency of administration).

It is also desirable to combine this peptide with GHRP-6 or GHRP-2. For example, the standard protocol is as follows: one injection contains 100 µg of GHRP-6 + 100 µg of modified GRF(1-29). You can dissolve it in one syringe. The solution is administered 2-3 times a day, at least 3 hours apart. The best time for injections is after training, 15 minutes before meals, and before going to bed.
